Mystery to the origin of long-lived, skin-deep immune cells uncovered
ScienceDaily (June 7, 2012) ? Scientists at A*STAR's Singapore Immunology Network (SIgN) uncovered the origin of a group of skin-deep immune cells that act as the first line of defense against harmful germs and skin infections. SIgN scientists discovered that these sentry cells of the skin, called the Langerhans cells (LCs), originate from two distinct embryonic sites -- the early yolk sac and the fetal liver.
LCs are dendritic cells (DCs) found in the outermost layer of the skin. DCs are a critical component of the immune system because they are the only cells able to 'see' and 'alert' other responding immune cells to initiate a protective response against harmful foreign invaders. Like sentries of the immune system, DCs are strategically positioned where they are likely to encounter harmful pathogens. Identifying the source of these specialized immune cells may hold exciting possibilities to novel strategies for vaccination and treatment of autoimmune diseases and inflammatory skin disorders.
In contrast to other DCs which are constantly replaced by a circulating pool of bone marrow-derived precursors, LCs has the interesting ability to maintain themselves throughout life. While it is established that these long-lived sentry cells of the skin arise from precursors that are recruited to the skin prior to birth, this is the first time that the exact origin of the precursors of LCs is revealed through advanced fate-mapping technique (a method of tracing cell lineages to their embryonic origin).
In this study, published in the June issue of Journal of Experimental Medicine, Dr Florent Ginhoux, and his team demonstrated that adult LCs originate from two distinct embryonic lineages in two succeeding waves. The first wave of precursor cells from the yolk sac 'seed' the skin before the onset of the fetal liver. Interestingly, the team discovered that at the later stage of development, the yolk-sac precursors are largely replaced by a type of white blood cells from the fetal liver.
Said Dr Ginhoux, Principal Investigator of SIgN, "Whether this unique dual origin of Langerhans cells influences their ability to maintain skin integrity or dictate their specialized immune functions in response to microbes and vaccines needs to be examined. But having identified their origin surely opens new possibilities of using them as novel vaccination strategies or as therapeutic tool for treating inflammatory skin diseases like psoriasis."
Scientific Director of SIgN, Professor Paola Castagnoli said, "This discovery sheds light on understanding the complexities of the immune system, in particular the relationship between immune responses and human diseases. It will bring us closer to our goal of discovering novel ways of treating and preventing a range of immune diseases that will impact healthcare."

Smoak HRs twice, M's crush Rangers 21-8

ARLINGTON, Texas (AP) - Justin Smoak hit two three-run homers against his former team and the Seattle Mariners used a pair of early eight-run innings to rout the Texas Rangers for the second straight game, winning 21-8 Wednesday night.
Smoak had three hits to go with his career high for RBIs as Seattle reached double digits in back-to-back games against the AL West leaders. Jesus Montero homered among his three hits and drove in four runs and Kyle Seager had four hits and two RBIs. Every Seattle starter had at least one hit in amassing 20 hits.
Dustin Ackley also had a three-run homer in support of former Rangers minor league prospect Blake Beavan (3-4).
Rangers starter Derek Holland (4-4) gave up eight hits, eight runs and two walks in 1 2-3 innings Seattle had lost five in a row before beating Texas 10-3 Tuesday night. They scored eight runs in the second and third innings en rout to scoring the second-most runs in club history, equaling a 21-9 victory against the Los Angeles Angels on Sept. 30, 2000.
Coincidentally, that was the same day the Rangers gave up the most runs in club history, a 23-2 loss at Oakland.
The Mariners started the game with only one player hitting better than .250 in the lineup. No matter, they sent 27 batters to the plate in the second and third innings, scoring 16 runs on 15 hits - tying their season high for a game - including three home runs.
And they did it without Ichiro Suzuki, who was held out by manager Eric Wedge to get some rest.
The 21 runs are the most scored in the majors this season, topping the Rangers and Rockies, who have each scored 18 runs.
Holland got off to a quick start, retiring the top of the first in order, including strikeouts of Chone Figgins and Kyle Seager, the best hitter by average in the Mariners lineup on Wednesday at .265.
The Mariners found their groove in the second, sending 13 batters to the plate. Ackley and Montero each homered and accounted for five RBIs, all with two outs, in the inning.
Miguel Olivo had two RBIs in the inning, and Seager had one.
Only Michael Saunders did not reach safely in the inning, though he made up for that with a double and a run scored to start the Mariners' encore, an eight-run third inning.
Montero had two more RBIs and Smoak added a three-run home run.
The Rangers needed 97 pitches to retire six hitters.
Holland, who was 6-1 in seven career starts against Seattle, including 4-0 with a 2.70 ERA since the start of last season, threw 41 pitches in the second inning before being relieved by Yoshinori Tateyama.
Tateyama retired the final out of the second, but recorded only one out in the third, giving up seven hits and six earned runs.
The Mariners added four more in the eighth. Ackley reached and scored on Seager's double, his fourth hit, and Montero walked for the second time before Smoak hit his second home run.
The Rangers tried to make a game of it, sending nine men to the plate in scoring five runs in the sixth. Texas added two in the seventh.
Outfielder Josh Hamilton, who was back in the lineup after missing the previous two games with a respiratory illness, was 2 for 4 with two doubles, one a run-scoring hit that increased his major league-leading RBI total to 57.
NOTES: Hamilton has hit safely in 40 of 47 games this season. Rangers manager Ron Washington gave shortstop Elvis Andrus the night off. Andrus acknowledged the time off was a mental break after he committed an error on Tuesday. Cowboys quarterback Tony Romo threw out the ceremonial first pitch - high but across the plate - caught by Rangers reliever Mike Adams, a Cowboys season-ticket holder.

SPRING Singapore?s Enterprise Development Centres?are stepping up their efforts to help SMEs stay?competitive and productive in uncertain economic times.
By partnering trade associations and chambers (TACs), SPRING?Singapore aims to reach out to 20,000 enterprises annually and?help them capitalise on grant schemes to raise productivity. This?marks an increase from the 14,200 firms that received productivity?and business advice from the EDCs last year.
?We recognise that SMEs need help to make this transition to?create innovative products and services, upgrade skills, design?better jobs, leverage technology and find new markets overseas,??says Mr Teo Ser Luck, Minister of State for Trade and Industry, at?the fourth EDC Conference on 28 March this year.
SMEs can approach any of the EDCs, which are located at the?premises of five TACs, for one-to-one advisory services on areas?such as financial and human resource management, branding,?and overseas expansion.
?Companies today need to enhance their competitiveness in the?marketplace and keep pace with rapid changes in technological?innovation, market upheavals and changing consumer needs so?as to ensure their survival and growth,? says Mr Hernaikh Singh,?director of the EDC at the Singapore Indian Chamber of Commerce?& Industry.
?The EDCs play an important role in helping companies identify?and implement relevant assistance programmes to address their?needs,? he adds.

a manpower shortage. We knew the only way we could increase?our capacity was with existing or fewer headcount. One option?was to automate our laundry processes but we didn?t know?how.? Acting on the advice provided by business advisors at the?Singapore Manufacturers Federation, the company embarked?on the Productivity Management Programme. Following an?in-depth diagnosis of the company?s productivity issues, Orchid?Laundry was able to streamline and automate its processes to?improve its performance.
For family-owned food manufacturer Hock Lian Huat Foodstuff,?process automation helped it improve its productivity. The company,?which approached the EDC at the Singapore Manufacturers??Federation for productivity advice, invested in a new machine?to make prawn crackers, an ingredient in ?ngoh hiang?, the?traditional meat rolls that are one of its main products. Now?only two workers are needed for this process, down from four.
EDCs provide one-to-one advisory services on?areas such as financial and human resource?management, branding, and overseas expansion.
The EDC also helped the company to tap $120,000 of tax rebates?under the Productivity and Innovation Credit (PIC) scheme. To?help more SMEs understand how they can benefit from the PIC?programme, the EDCs will conduct a new series of clinics from?May to October this year. About 500 SMEs are expected to benefit?from these sessions. The clinics, which are run in collaboration?with the Inland Revenue Authority of Singapore (IRAS), benefited?some 300 SMEs last year.
